Alzheimer’s Disease beyond Calcium Dysregulation: The Complex Interplay between Calmodulin, Calmodulin-Binding Proteins and Amyloid Beta from Disease Onset through Progression

A multifactorial syndrome, Alzheimer’s disease is the main cause of dementia, but there no existing therapy to prevent it or stop its progression. One earliest events disruption calcium homeostasis that just a prelude disease’s devastating impact. Calcium does not work alone must interact with downstream cellular components which small regulatory protein calmodulin central, if primary. This review supports idea that, due dyshomeostasis, dominant functions in all stages disease, and these are impacted by amyloid beta. Amyloid beta only binds regulates also multiple calmodulin-binding proteins involved Alzheimer’s. Together, they act on regulation neuroinflammation, amyloidogenesis, memory formation, neuronal plasticity more. The complex interactions between calmodulin, binding may explain why many therapies have failed doomed failure unless considered.
